Infected w/ Rootkit.Tdss...Need help

Hi guys, I've been reading posts in the forums and have downloaded Mbam, but when I double click to open the file the box just disappears and nothing happens...I currently run threatfire as my only virus software.

The main problem is I have been infected with a few differant viruses but the nastiest is rootkit.tdss....Any help or suggestions would be greatly appreciated! I use this computer atleast 8 hours a day as I work from home, so this virus is really messing getting annoying and only getting worse.
