Possible virus taking over system

I'm a novice with viruses and quickly losing administrative rights on single-user, stand-alone system: normal boot fails; user rights and major programs blocked (Security, Installer, System Restore, Backups, CD/DVD, printer, etc.); driver updates and downloads fail; Avast! Resident scanner disappeared, although full scans reactivated. I'm losing system functions quickly, so desparately need help.

Thank you for the prompt response - however, MBAM won't update; only scans. Are there any suggestions to force the update?

Do you get an error message? Please give all the details you can so that I may assist in the fastest and best way possible. Try again, I had mine give an error message earlier I think there may have been a website glitch.

Mbam message: "Update failed. Make sure you are connected to the Internet and your firewall is set to allow Malwarebytes' Anti-Malware to access the internet." Internet connection is active and I added Malwarebytes.org to IE options > trusted sites, since firewall is inaccessible, but update still failed. Even tried downloading to program, but mbam doesn't recognize the file.
