As for the Malwarebytes update and scanner settings, its up to you. Typically there are multiple updates a day. So you could choose hourly (Malwarebytes will check for updates hourly), realtime (will be updated as they come out), daily (Malwarebytes will check once a day for updates at the time you choose), or one of the other options.

As for the scanner, since the quick scan does not take much time, many people choose to scan daily. Others scan once a week or a couple times a week. It's all up to you.

My personal preference is manual Malwarebytes updates and manual (once a week or a couple times a week) scans.

I have had NIS 2009 upgraded to 2010 and MBAM pro for a nearly two years now and never a problem. Vista 32 bit OS. I have not changed the default configuration that MBAM came with.
