Running Windows Defender

Yes you may.  Malwarebytes is designed to be able to run in real-time alongside other antivirus/anti-malware applications so that customers may use these as additional layers of defense if they wish to.  In order to enable Windows Defender with Malwarebytes Premium installed, assuming it isn't already (by default Malwarebytes checks to see if Windows Defender is turned on, and if it is, it leaves it turned on), just open Malwarebytes and navigate to Settings>Application and under the Windows Action Center section select the option Never register Malwarebytes in the Windows Action Center, then restart your system and Windows Defender should be enabled from that point on.  If not then you may need to check your settings in Windows Defender to enable it.
