First of all I'm thinking about buying the PRO version, but is the licence duration only 1 year or for life time?

And secondly do I still need Microsoft Security Essentials, when I have the PRO-version?

Yes, you still need MSE or another free or paid anti-virus, as MBAM is not an anti-virus (AV). Rather it is designed and tested to run alongside a robust AV, to provide layered, complementary protection against zero-day threats often missed by standard AVs. Does Malwarebytes Anti-Malware replace antivirus software?
